Taenia solium causes two diseases in humans, cysticercosis and taeniosis. Tapeworm carriers are the main risk factor for neurocysticercosis. Limited information is available about the immune response elicited by the adult parasite, particularly the induction of Th2 responses, frequently associated to helminth infections. Calreticulin is a ubiquitous, multifunctional protein involved in cellular calcium homeostasis, which has been suggested to play a role in the regulation of immune responses. In this work, we assessed the effect of recombinant T. solium calreticulin (rTsCRT) on the cytokine, humoral and cellular responses upon experimental infection in Syrian Golden hamsters (Mesocricetus auratus). Animals were infected with T. solium cysticerci and euthanized at different times after infection. Specific serum antibodies, proliferative responses in mesenteric lymph nodes and spleen cells, as well as cytokines messenger RNA (mRNA) were analyzed. The results showed that one third of the infected animals elicited anti-rTsCRT IgG antibodies. Interestingly, mesenteric lymph node (MLN) cells from either infected or non-infected animals did not proliferate upon in vitro stimulation with rTsCRT. Additionally, stimulation with a tapeworm crude extract resulted in increased expression of IL-4 and IL-5 mRNA. Upon stimulation, rTsCRT increased the expression levels of IL-10 in spleen and MLN cells from uninfected and infected hamsters. The results showed that rTsCRT favors a Th2-biased immune response characterized by the induction of IL-10 in mucosal and systemic lymphoid organs. Here we provide the first data on the cytokine, antibody and cellular responses to rTsCRT upon in vitro stimulation during taeniasis.  相似文献

Cryosurgery with liquid nitrogen is one of the most used treatments for actinic keratosis. We aimed to study the effectiveness of two consecutive sessions of cryosurgery for actinic keratosis and investigate factors associated with its therapeutic success. Hence, we conducted a longitudinal study including 92 patients of both sexes, aged 50–75 years with 5–50 actinic keratosis on the face and forearms, who underwent cryosurgery and treatment with sunscreen SPF 30, at baseline and after 120 days. The lesions were counted in duplicate by the same examiner before the start of treatment and after 120 (N = 92) and 300 days (N = 33), represented by their medians and quartiles and compared using the generalized linear mixed effects model (negative binomial). Treatment behavior was investigated in relation to sex, age, education, skin type, smoking, sun exposure at work and the use of aspirin, anti-inflammatory and angiotensin-converting enzyme inhibitors. There was a significant reduction in the actinic keratosis count on the face and forearms (p < 0.05). Our results confirmed the effectiveness of cryosurgery for actinic keratosis, with a 57% reduction in the number, and size of the lesions. Higher education levels (p = 0.02) and less sun exposure at work (p = 0.02) independently promoted a significant reduction in the actinic keratosis count. Different population groups showed characteristic responses to the treatment, which may be explained by the degree of adherence to the use of photoprotection. In two sessions, cryosurgery with liquid nitrogen reduced the actinic keratosis count.  相似文献

Leishmania (V) braziliensis is one of the most important ethiologic agents of the two distinct forms of American tegumentary leishmaniasis (cutaneous and mucosal). The drugs of choice used in leishmaniasis therapy are significantly toxic, expensive and are associated with frequent refractory infections. Among the promising new targets for anti-protozoan chemotherapy are the proteases. In this study, serine proteases were partially purified from aqueous, detergent and extracellular extracts of Leishmania braziliensis promastigotes by aprotinin-agarose affinity chromatography. By zymography, the enzymes purified from the aqueous extract showed apparent activity bands of 60 kDa and 45 kDa; of 130 kDa, 83 kDa, 74 kDa and 30 kDa from the detergent extract; and of 62 kDa, 59 kDa, 57 kDa, 49 kDa and 35 kDa from the extracellular extract. All purified proteases exhibited esterase activity against Nalpha-benzoyl-L-arginine ethyl ester hydrochloride and Nalpha-p-tosyl-L-arginine methyl ester hydrochloride (serine protease substrates) and optimal activity at pH 8. 0. Proteases purified from the aqueous and extracellular extracts were effectively inhibited by benzamidine (trypsin inhibitor) and those from the detergent extract were inhibited by N-tosyl-L-phenyl-alanine chloromethyl ketone (chymotrypsin inhibitor) indicating that all these enzymes are serine proteases. These findings indicate that L. braziliensis serine proteases display some biochemical similarities with L. amazonensis serine proteases, demonstrating a conservation of this enzymatic class in the Leishmania genus. This is the first study to report the purification of a serine protease from Leishmania braziliensis.  相似文献

Named after the Sanskrit word netr, which means 'one who guides', the netrin family of secreted proteins provides migrational cues in the developing central nervous system. Recently, netrins have also been shown to regulate diverse processes (such as cell adhesion, motility, proliferation, differentiation and, ultimately, cell survival) in a number of non-neuronal tissues. In some cases, netrins affect these functions through non-classic netrin receptors, prompting a renewed interest in these factors in and beyond the nervous system.  相似文献

Biological catalysis involves interactions distant from the site of chemistry that can position the substrate for reaction. Catalysis of RNA 2′-O-transphosphorylation by the hepatitis delta virus (HDV) ribozyme is sensitive to the identity of the N(–1) nucleotide flanking the reactive phosphoryl group. However, the interactions that affect the conformation of this position, and in turn the 2′O nucleophile, are unclear. Here, we describe the application of multiple substrate internal competition kinetic analyses to understand how the N(–1) nucleobase contributes to HDV catalysis and test the utility of this approach for RNA structure–function studies. Internal competition reactions containing all four substrate sequence variants at the N(–1) position in reactions using ribozyme active site mutations at A77 and A78 were used to test a proposed base-pairing interaction. Mutants A78U, A78G, and A79G retain significant catalytic activity but do not alter the specificity for the N(–1) nucleobase. Effects of nucleobase analog substitutions at N(–1) indicate that U is preferred due to the ability to donate an H-bond in the Watson–Crick face and avoid minor groove steric clash. The results provide information essential for evaluating models of the HDV active site and illustrate multiple substrate kinetic analyses as a practical approach for characterizing structure–function relationships in RNA reactions.  相似文献

Plantations are frequently established on abandoned pasture lands to speed forest recovery. This strategy requires matching a tree species mix with the prevailing microenvironmental conditions. In four degraded pastures of the Mexican Lacandon rainforest, we planted 2,400 trees of 6 species (Guazuma ulmifolia, Inga vera, Ochroma pyramidale, Trichospermum mexicanum, Bursera simaruba, and Spondias mombin) to (1) test survival, initial growth, and establishment costs; (2) evaluate whether vegetative cuttings outperform direct seeding or transplants of nursery‐raised seedlings; (3) determine tree response to herbaceous dominance and soil compaction; and (4) scrutinize the results' consistency across sites and sampling scales of tree–microenvironment interactions (individual tree vs. averaged plot responses). After 2 years, overall survival and growth rates were high for 2 of 3 nursery‐raised species. Contrary to expectations, all seedlings outperformed the cuttings while direct seeding resulted in a cost‐effective option of intermediate efficacy. The impact of soil resistance to root penetration on tree biomass accumulation was species dependent while bulk density was not relevant. Soil‐covering, herbaceous vegetation accelerated growth in 3 of 4 tested species during the dry season. At this initial stage of forest restoration in abandoned pastures, Guazuma and Trichospermum were the most restoration‐effective species. Costs can be reduced by using direct‐seeding Inga and avoiding weeding during the dry season. Finally, our results demonstrate how species selection trials can be misleading due to site variations in tree response and to sampling scales that fail to account for small‐scale environmental heterogeneity. We recommend ways to improve the design of restoration trials.  相似文献

This study assessed the presence of cleaved caspase 3 (CC3) during the in vitro development of swine embryos produced by parthenogenetic activation (PA). Embryos with high and low capacity to develop into blastocysts and the exposure to a caspase inhibitor (z‐DEVD‐fmk) were used to investigate the effect of CC3 on embryo development. The blastocyst rate (64.3% vs. 16.4%) and the average number of nuclei per blastocyst (39.7 vs. 19.8) were significantly higher (P < 0.05) in early‐ (before 24 hr) compared to late‐ (between 24 and 48 hr) cleaving embryos after PA. CC3 was mainly detected in the cytoplasm of Day‐2 and ‐4 embryos, but was primarily localized in the nucleus of Day‐5 and ‐6 embryos. The fluorescence signal for CC3 relative to negative controls was significantly higher (P < 0.05) in early‐ (2.42‐fold) compared to late‐cleaving (1.39‐fold) embryos at Day 2 of culture. Treatment with z‐DEVD‐fmk during the first 24 or 48 hr of the culture period resulted in more embryos developing into blastocysts compared to the control group (55.8% and 55.1% vs. 37%, respectively; P < 0.05). This study confirmed the presence of CC3 in PA embryos from the two‐cell to the blastocyst stage, and revealed that CC3 cellular‐localization changed during embryo development. CC3 was shown to be more abundant in early‐cleaving and more developmentally competent embryos compared to late‐cleaving and less developmentally competent embryos.
